BtuGalA105B (unsaturated galacturonyl hydrolase)

BtuGalA105B

Exo-uGalA0224

(EC.3.2.1.173) unsaturated galacturonyl hydrolase

CAZy Family: GH105

PROPERTIES

1. ELECTROPHORETIC PURITY

-Single band on SDS-gel electrophoresis (MW ~46 kDa)

undefined

Figure 1. Electrophoresis analysis of BtuGalA105B. M, molecular weight marker (PageRuler Prestained Protein Ladder, Thermo Scientific); lane 1, culture lysate before IPTG induction; lane 2, culture lysate after IPTG induction; lane 3, BtuGalA105B purified from Ni sepharose fastflow column.

2. SPECIFIC ACTIVITY

Displayed activity on uGalAn (n=2 and 3) which was visualized by TLC. A complete loss of the uGalA2 is accompanied by the presence of a new GalA band.

.

3. RELATIVE RATES OF HYDROLYSIS OF SUBSTRATES

undefined

Figure 2. Profiles of oligogalacturonide digestion by GH105 (BtHgl105B).

4. PHYSICOCHEMICAL PROPERTIES

pH Optima: suggested use 4.0

Temperature Optima: suggested use 37°C

5. STORAGE CONDITIONS

The enzyme should be stored at -20°C. For assay, this enzyme should be diluted in 20 mM acetate (pH4.0). Swirl to mix the enzyme immediately prior to use.
